Intelligent light incubator research and analysis of the effects of light on the growth of Datura stramonium

July 06, 2023
There are a lot of Datura in Yangling, Shaanxi, and Datura has green stalks, white flowers of Datura, purple stems, and purple Datura Datura. In addition to these differences in appearance color, Datura and The differences in other biological characteristics of Datura stramonium are unknown. In particular, the cause of purple changes in purple Datura has not been reported. One of the necessary environmental factors for the growth and development of plants is light, which is treated with light intensity in an intelligent light incubator to study the growth and development of Datura and the relative microstructure of leaves and stems and leaves of Datura under different light intensities. The changes aim to clarify the influence of different light on the growth characteristics of Datura stramonium, explore the photoresponse mechanism of Datura stramonium evolution, and compare the growth characteristics of Datura stramonium and Datura stramonium at different light intensities. It provides a theoretical basis for the difference between the two, and has a theoretical significance for exploring the adaptive evolution mechanism of plant varieties.

In March 2010, the seed coats were peeled and germinated in a petri dish with a diameter of 11 cm. Each dish had 50 seeds. When the germination rate per dish reached 50%, seeds of similar bud length were sowed in pots with a diameter of 15 cm for intelligent illumination. The incubators were incubated in batches. The light intensities in each batch were weak light (5000 and 9000 lx), medium light (13000 and 18000 lx), and strong light (22000 lx). Each batch of Datura and Datura mandrilla cultivated 6 pots each. , 6C7 strains per pot. Each batch of culture parameters was light:dark=12h:12h, 25°C/20°C. Each index was measured after 50 days. Determination of morphological indicators: Twenty-two Drosophila individuals were sampled on the 50th day of culture. The plant height was measured with a small steel ruler, the base stem diameter was measured with a vernier caliper, and the number of leaves and bud length were counted, followed by dryness, 1/10000 balance. Measure dry weight. The leaf areas of the 3rd and 4th leaves of each Datura and Datura mandala were measured using a leaf area meter (Li-3100C, Li-COR, Lincoln, USA), and the leaves were dried and dried to measure the dryness by a 1/10000 balance. Heavy, record data, take the average.

The growth of Datura stramonium was observed in an intelligent light incubator. It was found that the Datura had the best growth under moderate light intensity, and weaker or stronger light was not conducive to its growth. The light intensity was in direct proportion to the leaf thickness, palisade tissue, sponge tissue thickness, stomatal density and stomatal index of Datura stramonium. The light intensity had an inducing effect on the anthocyanin in the stem of Datura stramonium, and the purple variation of Datura mandil was inferred. May be related to changes in light intensity.
